Madrigal Pharmaceuticals shares climbed 8.88% to $469.45 in Tuesday trading, as of 10:13 a.m. ET, adding $885 million in market value after the company reported its first profitable quarter. Sales of its liver drug Rezdiffra reached $431.3 million, up from $287.3 million a year earlier.

From loss to $55.4 million profit

The third-quarter results showed profit of $55.4 million, or $1.83 a share, a swing from a loss in the year-earlier quarter. Cash and securities stood at $961.8 million at the end of the quarter, the company said.

Rezdiffra has brought in $1.1 billion so far this year, a total Madrigal described as blockbuster status. As of Sept. 30, more than 55,850 patients were using the drug, roughly twice the prior-year count.

If the gain holds, it would be Madrigal's biggest one-day rise since March 26, when the shares rose 12.3%.

A drugmaker built on one pill

Madrigal develops treatments for MASH, a serious liver disease that can lead to cirrhosis, liver failure and liver cancer. Rezdiffra, its only approved product, is a once-daily pill for adults with MASH and moderate to advanced liver scarring. It has been sold in the US since April 2024 and in Germany since September 2025.

The company booked $958.4 million in Rezdiffra sales in 2025, according to its annual report. It also holds licenses to other MASH candidates, including an oral GLP-1 drug, ervogastat and six siRNA programs.

A rebound after a long slide

Before Tuesday, the stock was down about 26% for the year as of Monday's close, while the S&P 500 was up about 10%. It had fallen 14% over the prior month and sat 29% below its highest close of the past year, set on Dec. 22, 2025.

On the earnings call, management said it is starting a second cirrhosis trial of 2,000 patients, with a first readout pinned to late 2027.

Madrigal's market value was $10.8 billion in Tuesday's morning trading.
