Stoke Therapeutics, which develops RNA-based medicines for severe genetic diseases, reported a [third-quarter net loss of $73.6 million](https://www.sec.gov/Archives/edgar/data/0001623526/000119312526437775/stok-ex99%5F1.htm?ref=bullished.co), or $1.08 per share, up from $38.3 million, or $0.65 per share, a year earlier. The company said Thursday that its Phase 3 Dravet syndrome trial remains on track for a readout in the third quarter of 2027, followed by a planned U.S. approval filing in the second half of that year, according to an [8-K](https://www.sec.gov/Archives/edgar/data/0001623526/000119312526437775/stok-20261029.htm?ref=bullished.co).

## Spending on zorevunersen drives the loss

Research and development spending reached $59.8 million, from $37.7 million a year earlier. The company said the increase came from more activity and staff for zorevunersen, its lead drug for Dravet syndrome, a severe genetic epilepsy.

Sales, general and administrative costs rose to $28.7 million from $16.0 million, which the company attributed to staffing and launch readiness. The quarterly loss included $11.0 million of non-cash stock-based compensation, against $8.0 million a year earlier.

Quarterly revenue edged up to $11.3 million from $10.6 million. For the first nine months, revenue fell to $26.8 million from $183.0 million, because the 2025 period included $150.8 million tied to the Biogen license. The nine-month net loss was $185.3 million, against net income of $51.0 million a year earlier.

Cash, cash equivalents and marketable securities totaled [$358.8 million](https://www.businesswire.com/news/home/20261029090140/en/Stoke-Therapeutics-Announces-Third-Quarter-2026-Financial-Results-and-Provides-Business-Updates?ref=bullished.co) on September 30, which the company expects to fund operations into early 2028.

## Trial progress and the FDA meeting

Stoke uses antisense oligonucleotides, short strands of genetic material meant to help genes make more protein. Zorevunersen is in the Phase 3 EMPEROR study, which enrolled [162 patients](https://www.businesswire.com/news/home/20261029090140/en/Stoke-Therapeutics-Announces-Third-Quarter-2026-Financial-Results-and-Provides-Business-Updates?ref=bullished.co) in the U.S., U.K. and Japan by June 2026\. Biogen holds rest-of-world rights, while Stoke keeps the United States, Canada and Mexico.

CEO Ian F. Smith said about 110 of the 162 patients have passed Week 28, when the main goal of the trial, change in major motor seizure frequency, is measured. About 20 have finished Week 52 and are continuing in the open-label period. "To date, no patients have discontinued treatment," he said.

Stoke also met recently with the FDA about its planned application. For the cognition and behavior endpoint, it will use a combined score across four Vineland-3 subdomains: expressive communication, receptive communication, interpersonal relationships and personal skills. The company plans to submit data to the FDA throughout the first half of 2027 and is preparing for a potential U.S. approval and launch in early 2028.

## Eye disease program reads out next year

Stoke's second candidate, STK-002, is in the Phase 1 OSPREY study for autosomal dominant optic atrophy, an inherited eye disease. Dosing of the third cohort is underway, with the fourth expected in early 2027.

Initial safety and efficacy data from the STK-002 study are anticipated in the first half of 2027, ahead of the Dravet readout in the third quarter.
