He also said his scientific expectation is for efficacy in the ballpark of Dupixent's, with Phase 2b data due before year-end. ## A market sitting on the sidelines Mainolfi said published reports put the US population aged 12 and older with moderate-to-severe atopic dermatitis at about 6 million to 9 million. Only an estimated 400,000 receive advanced systemic therapies, about 5%. Kymera's market research, he said, shows many patients do not feel their disease justifies an injectable biologic or a drug with a box warning. Patients and physicians want a safe oral option, and the efficacy it needs, he said, "does not necessarily have to match biologics." He pointed to Otezla-style precedent as transcribed, saying a drug launched more than a decade ago with roughly half the efficacy of biologics became a multibillion-dollar franchise. He also cited Icotide in psoriasis, where he said 60% of new prescriptions go to new patients, "pretty impressive" for a market built over almost 15 years. ## Dupixent ballpark, with caveats A Deutsche Bank analyst asked how KT621 might compare with Dupixent at 16 weeks. Mainolfi said cross-trial comparisons are "technically not doable" and carry "an inherent flaw," and that he was defining the ballpark "unscientifically." He said blocking the same upstream signal with a STAT6 degrader appears to shut down downstream signaling similarly, and that what matters is delivering a commercially successful profile. He expects conjunctivitis to show up in the data, since it fits the biology. The 16-week placebo-controlled portion of the BROADEN2 study is complete, he said, and 98% of eligible patients enrolled in the 52-week extension. Kymera plans to start a Phase 3 atopic dermatitis study by mid-2027, assuming supportive data and alignment with the FDA.
